Project introduction
An aptamer is a single stranded oligonucleotide (RNA or DNA) that specifically binds to proteins or other small molecules. Typically, aptamers are single-stranded oligonucleotides surfacing with nanoparticles via C-S, S-S, or amide bonds. However, it is difficult to maintain the inherent characteristics of targeted aptamers after chemical modification. On the other hand, the preparation cost of nucleic acid modified by covalent bond is high, and the chemical modification process causes the degeneration of nucleic acid. Here, we designed a novel nucleic acid aptamer, which can be combined with targeted aptamers by base complementary pairing principle. The hydrophobic layer is formed by GC chain loaded organic dye. Nano aptamers can mimic biometric principles to deliver siRNA and hydrophobic drugs to mitochondria. Nucleic acid is a biocompatibility compound. Nucleic acid with mitochondrial targeting properties is a world-leading drug transporter. Two Chinese invention patents have been authorized.
Performance index
Nano nucleic acid particles is less than 50 nm. It has the characteristics of nucleic acid recognition sites and mitochondrial aggregation. There is no cytotoxicity.
Applicable scope and market prospect
1. RNA delivery: With the development of gene therapy, the research and application of RNA in the field of tumor therapy has attracted people's attention. However, the use of nucleic acid therapeutics is limited by RNA's lack of ability to migrate to the site of drug action. At present, the domestic nucleic acid aptamer has been commercialized, and the nano nucleic acid aptamer system of this invention has a high degree of green preparation, good industrialization prospect, and is a good RNA transporter.
